Chemotherapy induced painful peripheral neuropathy (CIPPN) is a deleterious side effect of taxanes, a class of chemotherapy drugs used to treat cancer. Paclitaxel (PTX) is a widely used chemotherapy agent that induces neuropathic pain in cancer patients. The mechanisms underlying PTX induced CIPPN are still not fully understood. Extracellular vesicles (EVs) are involved in cell-to-cell communication and may be a potential target in CIPPN. Herein, we show that Schwann cell (SC) derived EVs from peripheral nerve change in response PTX treatment. Further, EVs isolated from PTX treated mice elicit cell signaling responses in sensory neurons that are associated with pain.
